Nike.com: launch time reported to be 7 a.m. pacific; you can sign up on their website to get a text when it is out. Last time it showed up right around 7. UK launch had a limit of 1 per customer. @Rich Stanbaugh reports that you can get a 15% coupon (unique code) by chatting with an agent. I have heard that they will not be on the SNKRS app, but worth checking the regular Nike app.
FleetFeet.com: New version shows up on their site; launch time TBD. EDIT: available
RunningWarehouse.com: launch time between 6-8 a.m. pacific. Limit 3 per customer. EDIT: on sale right after 6, fully sold out in about 10 minutes
Eastbay.com: available at 7 a.m. pacific; excluded from discounts.
Nike stores: will not have them, at least in Seattle.

Updates on sizing:
Women's sizing: normally for a unisex shoe you would go down 1.5 sizes, but for these recommendations are to go 2-2.5 sizes
Men's sizing: reports that the new version runs true to size, though some say go up half a size with the new version
Overall "snugger and narrower" than the previous version due to the Flyknit upper, though the Flyknit will stretch over time.

The next release of Nike Vaporflys is on Thursday, October 4th. Based on high demand in the last sale and growing hype (including a marathon world record) since then I would expect them to sell out quickly. However, there are ways to get them at list price even if you can't be online right at 10 a.m. eastern.
For those not familiar with the Vaporfly: yes, these do seem to make most runners faster. A New York Times analysis of Strava times found that they were around 4% faster on average, and 1% faster than the next best shoe. The Vaporflys have a different, bouncy feel than most running shoes that takes some getting used to. The shoes are not very durable, and have been reported to start losing their benefits at around 50-100 miles. At $250 list that makes them a shoe I would save for key races and a few shakeout runs. I have used them barefoot for a sprint tri, and while they worked they also rubbed skin off one of my heels, so I would probably wear socks with them for anything longer (your mileage may vary, and the new mesh upper may help there).
The shoes being released on the 4th have been updated to include a lighter, one-piece Flyknit upper, and are still priced at $250. This Runner's World article covers some of the changes. Sizing should be similar to the earlier Vaporflys, though it's possible the upper will make them feel smaller. The earlier thread on Vaporflys has experiences may of us had on sizing; in general I would go with your average running shoe size, leaning toward the smaller size if you are in between (if in doubt, buy both if you can and return one since it will be hard to get a different size later).
How to get them
Running stores: if you have a running store in your area carrying the Vaporfly, that may be the easiest route. After the last release centered on the Boston Marathon, one store in Boston was reported to have stock for several days. Nike stores also were well stocked.
Chicago Marathon expo: probably also will be well stocked, at least on Friday.
Nike.com: should have a lot of inventory but it will go fast (I would not expect popular sizes to last more than 20-30 minutes), and there are a few tricks to know. First, set up a Nike account in advance including your payment info, since you will be given a limited amount of time to check out. The Nike Plus free membership also gets you free, faster shipping. Second, for payment I would choose a card that does not tend to stop transactions with fraud alerts (I had my first attempts rejected by Chase with fraud alerts). Third, don't expect the inventory to show up on the existing Vaporfly page; this is where many people (including me initially) missed it in April. If you don't see the Vaporflys when they are supposed to go on sale, start looking in other areas of the site. In April Nike put the new color on a different Vaporfly page that required a new search to find. Fourth, try the app first. This is how I was eventually found them in April, and the checkout was faster than the website (assuming you are logged in and have already entered your info).
RunningWarehouse.com: probably the cheapest route since you can use our sponsor code (on the team discount page). But you might have to babysit the page for a while; in April the inventory didn't show up at the Nike on-sale time; and when it did it sold out quickly.
Eastbay.com: in April this was the easiest place to buy online; they had inventory of popular sizes well into the evening. Confirmations were slow to come which was a little worrying, but they delivered as expected.

I'm kind of clueless in al l this But how does the ZoomFly compare? Nike's website says it has the carbon plate as well - anyone try running in these? Still $160 but may be an option if the shoes go fast again - just curious? Appreciate this detail and maybe I can find myself in front of a computer on Thursday. Cheers.

@Daniel Ballin although the Zoom Fly looks similar and has some significant updates, reviews I've seen say the performance is still not on par with the Vaporfly. It might be interesting as a trainer with a similar feel to the Vaporfly (though $160 is a lot for a trainer).
@Daniel Ballin @Mike Westover I recently bought the Zoom Flys and Love them! Let's be very clear, I've been a Hoka Guy for years, but find I do most of my runs now in Nike. I think the Zoom Fly feels like a slightly heavier (they're still very light) and more durable (tougher sole and outer) version of the Vaporfly. The Vaporfly is great for races, but given their extreme lightness and rumored ~100 mile lifespan, they are NOT a training shoe for me. But I'm guessing I'll get at least a couple hundred (maybe more) miles out of the Zoom Fly's. I also have the Pegasus Turbo's and they are more expensive (can't figure out why) than the Zoom Fly but feel less like the Vaporfly (in my opinion). I still run in the Pegasus occasionally, but never get that "wow" feeling in them. I do get that feeling in the Zoom Fly. I'll be doing my races in the Vaporfly, my speed work and Threshold runs in my Zoom Fly, and mixing my random other runs between my Hoka Cliftons and my Zoom Flys.

Thanks @JohnWithrow I ordered a pair of ZoomFly yesterday from Running Warehouse with the EN discount so $136 for trainers. If I like them I'll jump on Thursday to get the Vaporfly's for racing. I typically run in Saucony kinvara, ISO freedoms and Altra's so I am used to 0-4mm drop. Will see how the 10mm drop goes. Thanks @MikeWestover for the tips for Thursday.
I can't comment at all on the 4% Vaperflys, but I like @Daniel Ballin have been a Saucony ISO freedom girl and now have 3 pair of Zoomfly's in the house. I read a review that they felt like the Clifton 1's. That's why I gave them a shot, not their close relation to the Vaperflys.
I feel FASTER in them. I noted it in all of my run entries. The legs just turn over easier. Then I did a little research and found they are related to the Vaperfly's. I got mine on sale AND with the EN discount from runningwarehouse. Sadly I burn through shoes (outer heel wear) in less than 175 miles on most shoes, besides the ISO's. Hokas, Kinvaras, etc. These will be no different.
The great news is I am not a fast runner. AT ALL. Many of the reviews were stating you have to be a 7:30 or faster mile pace to see benefits, but I don't feel that to be true at all. I will be racing IMKY in them. My ISO's feel so "dead" compared to them. I don't expect them to impart better fitness to my legs on the day, but I hope to keep my steady running steady for longer with the cushioning and faster feeling underfoot.

Anna Chatted with Nike from their website and they gave her a 15% coupon for the Vaporfly. I don' t think it is a sharable coupon, but you probably worth chatting and trying to get the coupon.
@scott dinhofer just the "Nike" app. You should be able to log in with your Nike Plus ID. You could try setting up a dummy order for another shoe to check it is good to go.
